It is specially adjusted ultrapure water DNase and RNase free. Usually, RNases are removed from H2O by the treatment with diethylpyrocarbonate (DEPC), which is then resolved by autoclaving. Degradation products of DEPC can negatively influence the course of RT-PCR and even PCR. In our product PCR Ultra H2O, RNases were removed by double affinite chromatography on special carrier. For this purpose PCR Ultra H2O does not contain any degradation products of DEPC and can be universally used for any work with RNA and in RT-PCR and PCR.

Plain Combi PPP Master Mix is a modification of Combi PPP Master mix (Cat. No. C208 - C210) not containing loading dye. This Mix is first choice for those applications where loading dye would interfere with measurements of DNA products e.g. by capillary chromatography or real-time PCR. The mix contains Taq DNA polymerase, deoxyribonucleotides, reaction buffer components, additives and anti-Taq DNA polymerase.

Taq DNA polymerase Unis is an alternative to Taq DNA polymerase (Cat. No. T032-T034). Difference between these two products is in difference in enzyme storage buffer which enhances stability and efficiency of the enzyme. Occasional amplification of nonspecific fragments can be prevented by enhanced dilution of the enzyme.

This product is a modification of Taq DNA polymerase Unis (Cat. No. T 037-T 039). Difference between these two products rely in lower concentration of the polymerase (1 U/µl) , which makes easier pipetting of the enzyme. Furthermore, the enzyme is supplied with 10x concentrated PCR Blue buffer. This buffer contains ammonium sulfate, which contributes to broader tolerance of MgCl2 and lower production of nonspecific enzymes.

Estimation of the size of DNA fragments generated in PCR (PCR fragments) or by DNA cutting by restriction enzymes (restriction fragments) is usually based on comparison of these fragments with DNA fragments of known size (DNA markers). Size of PCR fragments is often in the range 150-1000 base pairs. This range is covered by DNA fragments, which are part of DNA marker 155-970 (Fig. 1). These fragments were produced by cutting a reference plasmid by restriction enzyme. This marker DNA marker is supplemented innert dye (Purple) and additives increasing density and therefore can be loaded directly into agarose gel. This marker is especially suitable for analysis of PCR products generated by Taq-Purple DNA polymerase (Cat. No. T 107-T109). 

Aptamer Hot Start Master Mix is an optimized ready-to-use solution containing Taq DNA Polymerase, DNA aptamer anti-Tag, dNTPs, dye, MgCl2, additives and stabilizers. Additives and the dye allow direct loading of the PCR-amplified samples into the gel without adding loading buffer. When compared to hot start master mix containing Taq polymerase-specific monoclonal antibody, aptamer-based hot start Master Mix has an advantage that (1) it is more resistant to elevated temperatures, (2) the inhibitory effect is reversible, and (3) master mix is more chemically defined. The master mix is not recommended for applications in which fluorescent DNA probes are used for detection amplified DNA (qPCR). It is ideally suited for routine PCR amplification of DNA fragments up to 5 kb. It is compatible with template DNA extracted with DEP-25 DNA extraction kit.

Additives and the dye present in Combi PPP Master Mix allow direct loading of the PCR amplified samples into the gel without adding loading buffer. When compared to PPP Master Mix (Cat. No, P124 - P126), it contains a monoclonal antibody which binds to and inactivates enzymatic activity of Taq DNA polymerase. The master mix is not recommended for any applications in which fluorescence excitation is used, such as qPCR.

This product is the most advanced variant of Master mix for PCR.

PPP Master Mix without MgCl2 is similar to PPP Master Mix (cat. No. P124 - P126) but does not contain MgCl2, which is provided separately. This allows optimization of Mg2+ concentration, which is of key importance for proper PCR. The Mix contains Taq DNA polymerase, deoxyribonucleotides, reaction buffer components and additives.

When compared to antibody the aptamer has several advantages: (1) it is not irreversibly denatured at temperature upto 100oC, (2) has low molecular weight, is prepared synthetically, and has well defined composition, (3) is not contamined with components of mammalian cells, (4) is resistant to action of proteases and allows preparation of PCR mixes at room temperature, and (5) is less expensive.
